Context: Analyzing the neuro-immune mechanisms which result from persistent brain infection that may alter neuronal DNA damage response and their contribution to the progression of cognitive decline in Alzheimer’s disease. This project is part of the Occitania Region-sponsored INSPIRE initiative to shed light on mechanisms involved in aging, and to identify strategies to promote healthy aging. This project consists in analyzing the mechanisms underlying cognitive decline in Alzheimer’s disease (AD), using murine models of the disease. In this neurodegenerative and neuroinflammatory disease, we will explore the impact of the inflammatory response on the severity and the progression of AD-related symptoms. Moreover, we plan to analyze the consequences of infections by neurotropic pathogens, such as the parasite Toxoplasma gondii, on the evolution of AD. We found that several proinflammatory cytokines and neuronal infection by Toxoplasma gondii cause DNA double strand breaks to accumulate into the DNA of neurons. Finally, we will decipher what role these various processes may play in epigenetic.
